The distilled water method was stored at room temperature and 4°C respectively, and the -80°C freezing preservation used both the program-controlled cooling method and the foam box cooling method. After 20 months of storage, the preservation effects of the 5 strains preserved by the four different methods were compared. The experimental results show that the survival rate of the bacteria in the distilled water room temperature storage method is 100%, and the germination period is short, which is 4-10 days. It is a simple, practical, effective and low-cost long-term storage method; the survival rate of the -80°C freezing storage method is 56%-76%, and the germination period is 7-16 days. The foam box cooling method can well control the cooling speed and is a simple and effective temperature control method.
