Therefore, if the rubella virus IgG antibody titer is increased by more than 4 times in the duplicate sera, it is an indicator of recent rubella virus infection regardless of whether the IgM antibody is positive. How to operate (1) Sample collection: For screening tests, only a single blood sample is required. However, if the immune status of a person infected with the virus needs to be determined, samples must be taken from suspected rubella patients within 3 days after the onset of the rash and within the following 14 to 21 days for simultaneous testing. (2) As with general ELISA, add 50 μl of PBS to each well of the control and sample wells, followed by 10 μl of the sample, incubate at 25°C for 45 min, wash, and dry. (3) Add 250 μl of enzyme marker to each well and keep warm and wash in the same way. (4) Add 250 μl of pNPP substrate solution, keep warm and wash in the same way, then add 50 μl of 1 mol/L sodium hydroxide to terminate the reaction. Measure the absorbance of each well at 405 nm to determine the results of the tested samples. (5) If the result is positive, the sample can be further diluted and the antibody titer can be measured. The results of the two samples can be compared. Rubella virus is very harmful to children and can even cause fetal malformations in severe cases. If the result of the rubella virus test is positive, it means that you have the virus and it is not suitable for pregnancy at this time. You must first wait six months after the virus is completely cured before you can prepare for pregnancy. |
