(3) Add 250 μl of enzyme marker to each well and keep warm and wash in the same way. (4) Add 250 μl of pNPP substrate solution, keep warm and wash in the same way, then add 50 μl of 1 mol/L sodium hydroxide to terminate the reaction. Measure the absorbance of each well at 405 nm to determine the results of the tested samples. (5) If the result is positive, the sample can be further diluted and the antibody titer can be measured. The results of the two samples can be compared and a judgment can be made. determination 1. Both IgG and IgM antibodies to rubella virus are positive, or the IgG antibody titer is ≥1:512, indicating recent infection with rubella virus. 2. Both IgG and IgM antibodies of Rubella virus are negative, indicating that there has been no Rubella virus infection. 3. The IgG antibody titer of rubella virus is <1:512, and the IgM antibody is negative, indicating a history of infection. 4. In addition, reinfection with rubella virus is difficult to detect because IgM antibodies only appear briefly or at very low levels. Therefore, if the rubella virus IgG antibody titer is increased by more than 4 times in the duplicate sera, it is an indicator of recent rubella virus infection regardless of whether the IgM antibody is positive. The normal range for rubella virus is that the test results are all negative, so when the test results of female friends are negative, they can get pregnant normally. However, if the test results are positive, you cannot get pregnant because your body is already carrying the rubella virus, which will affect the health of the child. |
