Guishao Dihuang Wan - Drug Identification

(1) Take this product and observe it under a microscope: the gelatinized starch clumps are off-white. The irregular branched masses are colorless and dissolve in chloral hydrate solution; the hyphae are colorless or light brown, with a diameter of 4 to 6 μm. The thin-walled cells are spindle-shaped, with slightly thick walls and very fine oblique interlaced texture. The parenchyma is gray-brown to dark brown, the cells are mostly wrinkled, and contain brown nuclei. The epidermal cells of the pericarp are orange-yellow, polygonal in surface view, and the antibulbar walls are slightly bead-like thickened. Calcium oxalate needle crystal bundles exist in mucous cells, with a length of 80 to 240 μm and a needle crystal diameter of 2 to 8 μm. The cork cells are light red to slightly purple, with slightly thick walls. The thin-walled cells are round, with oval pits and integrated pit groups.

(2) Take 6g of this product as water-honey pills and grind them into powder; or take 9g of small or large honey pills, chop them into powder, add 3g of diatomaceous earth, and grind them evenly. Add 40 ml of ether, reflux at low temperature for 1 hour, filter, evaporate the filtrate, add 1 ml of ether to the residue to dissolve it, and use it as the test solution. Take another 1g of Angelica sinensis reference medicinal material, add 15ml of ether, and prepare the reference medicinal material solution in the same way. According to the thin layer chromatography method (Appendix Ⅵ B), 5 μl of each of the above two solutions were taken and spotted on the same silica gel G thin layer plate, and n-hexane-ethyl acetate (9:1) was used as the developing solvent. The plate was developed, taken out, dried, and examined under ultraviolet light (365 nm). In the chromatogram of the test sample, fluorescent spots of the same color appear at the corresponding positions in the chromatogram of the control medicinal material.
