Significance of pepsinogen detection

The main significance of testing pepsinogen is to determine whether it is normal. Because the fluctuation of pepsinogen will affect the normal digestive function and normal gastrointestinal function, it will cause considerable harm to the individual's body. Therefore, you must pay attention to it in normal times and choose the treatment method and testing method according to your own situation to determine the current pathological state.

Test method: Take 6 test tubes, accurately add 1 ml of reference solution to 3 of them, and 1 ml of test solution to the other 3, place them in a 37℃±0.5℃ water bath, keep warm for 5 minutes, accurately add 5 ml of hemoglobin solution preheated to 37℃±0.5℃, shake well, and accurately time it, and react in a 37℃±0.5℃ water bath for 10 minutes. Immediately add 5 ml of 5% trichloroacetic acid solution, shake well, filter, and take the filtrate for later use. Take another 2 test tubes, add 5 ml of hemoglobin test solution to each, and place in a 37℃±0.5℃ water bath to keep moist for 10 minutes.

Then accurately add 5 ml of 5% trichloroacetic acid solution, add 1 ml of the test solution to one tube and 1 ml of the above-mentioned hydrochloric acid solution to the other tube, shake well, filter, and take the filtrate as the blank control for the test sample and reference substance, respectively. Measure the absorbance at a wavelength of 275 nm according to the spectrophotometric method (Appendix IVA), calculate the average values ​​AS and A, and calculate according to the following formula.

A×Ws×n Protease activity per 1g (unit) = As×W×10×181.19 where AS is the average absorbance of the reference substance; A is the average absorbance of the test substance; Ws is the amount of tyrosine in each 1ml of the reference substance solution, μg; W is the sample size of the test substance, g; and n is the dilution multiple of the test substance.

Under the above conditions, the amount of enzyme that can catalyze the hydrolysis of hemoglobin to produce 1 μmol of tyrosine per minute is one unit of protease activity.

Laboratory Method Name: Determination of Pepsin - Pepsin Assay

Scope of application: This method uses the pepsin assay to determine the titer of pepsin.

This method is applicable to pepsin.

Principle: Accurately weigh an appropriate amount of this product, add hydrochloric acid solution to make a solution containing about 0.2-0.4 units per 1 mL, and determine the potency of pepsin according to the pepsin assay method.

3. Hydrochloric acid solution

Sample preparation: 1. Preparation of test solution

Accurately weigh an appropriate amount of this product and dilute it with hydrochloric acid solution to make a solution containing about 0.2-0.4 units per 1 mL as the test solution.

2. Preparation of reference solution

Accurately weigh an appropriate amount of tyrosine reference substance and dilute it with hydrochloric acid solution to make a solution containing about 0.5 mg per 1 mL as the reference substance solution.

Note: "Precision weighing" means that the weight should be accurate to one thousandth of the weighed weight. "Precision measurement" means that the accuracy of the measured volume should meet the accuracy requirements of the pipette for that volume in the national standard.
